Super Tetris 3 presents four distinct puzzle variations within a single Super Nintendo release. The foundational experience is Tetris Classic, offering standard and endless single player options.

Multiplayer is supported through a dedicated family mode for four participants and a battle format for two players. This competitive option extends to several of the included game types beyond the basic version.

Two other modes introduce specific rule sets. Sparkliss incorporates explosive block clearing within a fantasy themed setting, featuring both staged and puzzle based challenges. The most distinctive variant is Magicaliss, which introduces a color matching system where pieces can be rotated to select one of three hues. Clearing a line of a single color removes all blocks of that color from the playfield, with special grey and rainbow pieces adding further strategic layers to the familiar falling block formula.
