DK: King of Swing is structured around a central climbing mechanic where the player navigates Donkey Kong by using the L and R buttons to grip and swing between pegs. The primary objective involves retrieving stolen contest medallions from King K. Rool, following their theft from Cranky's Jungle Jam event.

Combat and interaction are integrated into this climbing system. Donkey Kong can perform a charged attack by simultaneously holding two pegs, or he can throw collected objects like bombs and rocks at enemies. This peg-based movement defines the majority of the in-game action.
