"Daffy Duck in Hollywood" is a side scrolling action game where the player controls Daffy Duck, who is acting as a detective. His client, Yosemite Sam, has had his golden movie awards stolen by the villainous Mad Professor Duckbrain, who demands a ransom.

The core gameplay involves navigating through various film set environments, which include western, horror, jungle, and science fiction themes. Progression requires dealing with hazards, with a specific mechanic involving the defusing of dynamite found within the scenes to prevent explosions.

Daffy's primary tool is a bubble gun, used to capture enemies inside bubbles which then float away, removing them. This weapon can be upgraded to fire different bubble types. Additionally, a companion character described as a fairy duckmother can be acquired to provide protective support during the adventure.
