Super Mario Bros. 3 is a side-scrolling platform game designed for one or two players. Its narrative involves Mario and Luigi journeying across the Mushroom World to stop Bowser and his seven Koopaling children, who have stolen magical wands and transformed the local kings into animals, subsequently kidnapping Princess Peach.

The experience is structured around eight distinct worlds, each with a unique environmental theme like deserts, ice, or sky levels, which dictates the hazards and visual design. Players navigate a world map to select stages, find hidden paths, and visit Toad Houses for bonus items. The core action involves running and jumping through levels filled with enemies and obstacles, with progression requiring the completion of standard stages, fortress castles, and airship battles.

A significant expansion from earlier titles comes from the variety of power-ups. Alongside returning items like the Super Mushroom and Fire Flower, new suits grant specialized abilities. These include the Super Leaf for tail attacks and brief flight, the Frog Suit for swimming, the Tanooki Suit for flight and a stone transformation, and the Hammer Suit for projectile attacks. The Starman provides temporary invincibility, while rare items like the Warp Whistle allow players to skip ahead.

Victory in airship stages involves defeating a Koopaling, which restores a transformed king and recovers a wand. The final objective is a confrontation with Bowser to rescue Princess Peach. In the two-player mode, control alternates between Mario and Luigi on the map, with opportunities for competitive bonus games.
